Landmark Brain Map Reveals How Gene Activity Changes Across Human Lifespan

Scientists have created one of the most detailed maps yet of gene activity in the human brain, offering new insight into how the brain changes from infancy through old age and how those changes may relate to neurological diseases.

The research focuses on the prefrontal cortex, a region of the brain involved in planning, decision-making, behaviour and emotional regulation. Researchers analysed gene activity at the level of individual brain cells, producing a large-scale dataset that includes samples from almost 1,500 people.

The project examined more than 6.3 million individual brain cells, including neurons as well as immune and vascular cells. By comparing samples from people at different ages and with different neurological conditions, scientists were able to identify patterns that change throughout the human lifespan.

One of the studies found that the prefrontal cortex goes through distinct periods of molecular change. Researchers identified rapid remodelling during early life, a more stable period during adulthood, and another phase of remodelling beginning at around the age of 65. Some of the later changes involved cells associated with immune activity, stress responses and circadian rhythms.

The wider dataset also includes information relevant to conditions including Alzheimer’s disease, Parkinson’s disease, schizophrenia and other neurological and psychiatric disorders. Researchers say the scale of the resource could help scientists investigate how genetic risk is connected to specific types of brain cells.

However, the findings do not mean that scientists can yet predict whether an individual will develop a particular disease. The researchers say additional brain regions will need to be studied to build a more complete picture of how the human brain changes with age and disease.

The studies were published in Nature on September 23, 2026, as part of a collection of research examining gene activity in the human brain.
